Semax vs Selank — What’s the Difference?
A detailed comparison of two leading Russian-developed neuropeptides and why researchers study them both together.
Semax and Selank are two of the most extensively researched neuropeptides to emerge from Russian neuropharmacology. Both were developed at the Institute of Molecular Genetics of the Russian Academy of Sciences and have been studied across cognitive enhancement, neuroprotection and anxiolytic research models. Despite superficial similarities, they operate through distinct mechanisms — which is exactly why researchers frequently study them in combination.
What is Semax?
Semax is a synthetic heptapeptide derived from the ACTH 4-7 fragment of adrenocorticotropic hormone, extended with the sequence Pro-Gly-Pro (PGP) to enhance stability and CNS penetration. It was developed in the 1980s as part of a Soviet research programme into cognitive enhancement compounds.
Semax’s primary research interest is its upregulation of Brain-Derived Neurotrophic Factor (BDNF) — the key growth factor for neuronal survival, plasticity, and the formation of new neural connections. It also modulates dopaminergic and serotonergic neurotransmission, making it a versatile tool in neurochemical research.
What is Selank?
Selank is a synthetic analogue of the endogenous immunopeptide Tuftsin — a tetrapeptide fragment of IgG that plays a role in immune modulation. The Pro-Gly-Pro (PGP) extension added to create Selank significantly increases its metabolic stability compared to native Tuftsin.
Selank’s primary research profile is anxiolytic — its effects on GABAergic neurotransmission have been studied in preclinical stress-response and stress response models. Unlike benzodiazepine-class compounds, Selank’s GABAergic modulation occurs without the sedative or dependence-associated effects seen with direct GABA agonists, making it an interesting research model.

Semax Research Applications
- BDNF Expression Studies: Semax is one of the most potent BDNF-upregulating compounds among research neuropeptides, making it a key tool in neuroplasticity research.
- Dopaminergic Research: Studies have examined Semax’s modulation of dopamine receptor expression and turnover in various brain region models.
- Neuroprotective Research: Research has explored Semax in models of ischaemic injury and oxidative stress in neural tissue.
- Serotonergic Pathway Studies: Semax’s interactions with the serotonergic system make it relevant to research into mood and cognitive processing.
Selank Research Applications
- Anxiolytic Mechanism Research: Selank’s GABAergic modulation without direct receptor agonism makes it a useful model for studying non-benzodiazepine preclinical stress-response pathways.
- Stress Response Models: Research has examined Selank’s effects on corticotropin-releasing factor and stress-related neurochemical changes.
- Immunomodulatory Research: Derived from Tuftsin, Selank retains some immunopeptide properties relevant to immune-brain axis research.
- Memory and Learning Models: Selank’s BDNF activity, though less pronounced than Semax’s, contributes to research into memory consolidation models.
Why Study Semax and Selank Together?
The rationale for the Semax/Selank composite is their complementary neurochemical profiles. Semax’s stimulating, BDNF-upregulating properties pair naturally with Selank’s calming, anxiolytic GABAergic effects — allowing simultaneous study of both pathways without the competing confounders that would arise from studying either in isolation alongside a non-selective compound.
This combination is particularly useful for researchers studying the relationship between cognitive enhancement and preclinical stress-response modulation — two processes that are neurologically interconnected through shared BDNF and serotonergic pathways.

Key Published Research
Semax
- Neurotrophic signalling (2006): Semax increased BDNF and TrkB receptor expression in the rat hippocampus, the basis for its reputation as a BDNF-modulating peptide.1
- Brain ischaemia (2014): Genome-wide analysis of a rat focal ischaemia model showed Semax shifted the expression of genes linked to immune and vascular responses.2
- Clinical stroke study (2018): A Russian clinical study evaluated Semax in patients at different stages of ischaemic stroke.3
Selank
- Enkephalinase inhibition (2001): Selank inhibited enkephalin-degrading enzymes, proposed as one mechanism of its anxiolytic activity.4
- Clinical anxiety study (2008): A Russian clinical study reported anxiolytic effects in patients with generalised anxiety disorder and neurasthenia.5
- GABAergic gene expression (2016): Selank altered the expression of genes involved in GABAergic neurotransmission in rat brain tissue.6
- Chronic stress model (2017): Selank enhanced diazepam’s anxiety-reducing effect in rats under unpredictable chronic mild stress.7
Regulatory & Research Status (2026)
- Semax — US FDA: Semax was removed from the FDA’s Category 2 compounding list in April 2026. In July 2026 the FDA’s advisory committee voted 8–5 (one abstention) to recommend it for the 503A bulks list for proposed cerebral ischaemia, migraine and trigeminal neuralgia uses. The vote is non-binding.
- Selank: Selank was not among the peptides reviewed by the FDA in 2026 and remains unapproved outside Russia.
Evidence Quality & Limitations
Most clinical data for both peptides appear in Russian-language journals, often from small or open-label studies by the developing institutes, and independent replication elsewhere is limited. Mechanistic claims such as BDNF up-regulation and GABAergic modulation come mainly from rodent studies. No published study has examined Semax and Selank together, so pairing them is based on their complementary profiles rather than on combination data.
