Compound Overview
Semax is a synthetic heptapeptide based on the ACTH(4-10) fragment (Met-Glu-His-Phe-Pro-Gly-Pro) with a C-terminal Pro-Gly-Pro extension that confers resistance to enzymatic degradation. It was originally developed at the Institute of Molecular Genetics, Russian Academy of Sciences, and has been the subject of preclinical and clinical research primarily in Russian and European scientific literature.
Unlike full-length ACTH, Semax does not interact with melanocortin receptors associated with adrenal steroidogenesis. Its pharmacological profile has been studied independently of the hypothalamic-pituitary-adrenal (HPA) axis.
Mechanism of Action
Neurotrophic Factor Expression: Preclinical studies have reported that Semax modulates the expression of brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) in rodent models. BDNF is a key regulator of synaptic plasticity, neuronal survival, and differentiation in neuroscience research.
Monoaminergic Modulation: Research in animal models has observed effects on dopaminergic, serotonergic, and noradrenergic neurotransmission following Semax administration, with regional specificity in the hippocampus and prefrontal cortex.
Neuropeptide Processing: Semax has been studied for its influence on endogenous neuropeptide processing enzymes, including carboxypeptidase E and prolyl endopeptidase, which play roles in neuropeptide maturation.
