Compound Overview
NAD+ (nicotinamide adenine dinucleotide, oxidised form) is a coenzyme present in all living cells. It serves as a central electron carrier in cellular redox metabolism, participating in hundreds of enzymatic reactions including those of glycolysis, the tricarboxylic acid (TCA) cycle, and oxidative phosphorylation.
NAD+ has become a major focus of biogerontology and metabolic research following the discovery that intracellular NAD+ levels decline with age in multiple model organisms. This decline has been linked to changes in sirtuin activity, PARP function, and mitochondrial bioenergetics in preclinical studies.
Biochemical Roles
Redox Cofactor: NAD+ functions as an electron acceptor in catabolic reactions (NAD+ to NADH), serving as a critical substrate for dehydrogenase enzymes throughout central carbon metabolism.
Sirtuin Substrate: NAD+ is the obligate co-substrate for sirtuin enzymes (SIRT1-7), a family of NAD+-dependent protein deacylases implicated in chromatin regulation, metabolic adaptation, and stress response in preclinical models.
PARP Substrate: Poly(ADP-ribose) polymerases (PARPs) consume NAD+ during DNA repair processes. PARP-mediated NAD+ consumption has been studied as a contributor to cellular NAD+ depletion under conditions of genotoxic stress.
CD38 Substrate: The ectoenzyme CD38, an NADase, has been identified as a major consumer of NAD+ in ageing tissues. CD38 activity is a subject of active investigation in NAD+ metabolism research.

Combined with NAD+ this is a game changing stack! My lab Rat is feeling 15years younger!
NAD+has been a game changer for me. I have using it for many months and the differenceis like no other. I had stuck with a different supplier for a long time but they seemed to have some difficulties, I don’t like change but after several failed “relaunches” it was time to find somewhere new. I was so worried, it’s alot of money, but the service was quick and I’ve now used the product and can confirm it’s as good! If I was to have one criticism it’s probably because I’m used to paying considerably more and getting big fancy boxes, this felt like things had been kept simple. But hey! The Pen is just as good and much cheaper than my previous! Will stay with midspeps!
