Compound Overview
KPV (Lysine-Proline-Valine) is a synthetic tripeptide corresponding to the C-terminal fragment (residues 11-13) of alpha-melanocyte-stimulating hormone (alpha-MSH). Unlike the full-length alpha-MSH tridecapeptide, KPV does not bind melanocortin receptors (MCRs), indicating its biological activity operates through a distinct, receptor-independent mechanism.
KPV has been studied in preclinical research models since the 1980s, with particular focus on NF-kB pathway modulation and inflammatory signalling cascades.
Mechanism of Action
NF-kB Pathway Modulation: In vitro studies have demonstrated that KPV can enter cells and directly interact with components of the NF-kB signalling cascade, including inhibition of IKK-beta phosphorylation and nuclear translocation of the p65 subunit. This mechanism has been characterised as melanocortin receptor-independent (Kannengiesser et al., 2008).
Intracellular Uptake: Research suggests KPV enters cells via peptide transporter PepT1 (SLC15A1), a proton-coupled oligopeptide transporter expressed in intestinal epithelium and other tissues. This transporter-mediated uptake has been studied as a model for oral peptide bioavailability.
