Compound Overview
CJC-1295 No DAC (Mod GRF 1-29) is a synthetic analogue of growth hormone-releasing hormone (GHRH), comprising the first 29 amino acids of GHRH with four amino acid substitutions to improve metabolic stability. The “No DAC” designation indicates this variant lacks the Drug Affinity Complex, resulting in a shorter half-life (~30 minutes) compared to the DAC-conjugated form.
Ipamorelin is a synthetic pentapeptide growth hormone secretagogue (GHS) that selectively activates the ghrelin/GHS receptor (GHS-R1a). It is characterised by high selectivity for growth hormone release relative to other pituitary hormones, a property that distinguishes it from earlier-generation secretagogues such as GHRP-6 and hexarelin.
The composite format combines both compounds at a 1:1 ratio for use in research protocols investigating synergistic GHRH/GHS receptor co-activation.
Mechanism of Action
CJC-1295 No DAC — GHRH Receptor (GHRH-R): Binds to and activates the GHRH receptor on anterior pituitary somatotroph cells. GHRH-R activation stimulates cAMP/PKA signalling, leading to pulsatile GH gene transcription and secretion in preclinical models.
Ipamorelin — GHS Receptor (GHS-R1a): Activates the growth hormone secretagogue receptor type 1a, a G-protein coupled receptor expressed in the pituitary and hypothalamus. GHS-R1a activation amplifies GH pulse amplitude through a complementary intracellular pathway (PLC/IP3/DAG) distinct from GHRH-R signalling.
In preclinical research, co-administration of GHRH analogues and GH secretagogues has been observed to produce synergistic amplification of GH pulsatility, exceeding the effect of either compound administered alone.
